T4 RNA連接酶催化RNA的5個磷酸和3個羥基之間形成磷酸二酯鍵。它最活躍的是密封缺口的雙鏈RNA，但它也會加入單鏈RNA的末端，可以連接RNA的3個羥基與DNA的5個磷酸形成雙鏈形式。酶需要ATP，除非底物的5端已預先腺苷化。
T4 RNA ligase catalyses the formation of a phosphodiester bond between a 5′ phosphate and 3′ hydroxyl of RNA. It is most active in sealing nicked double-stranded RNA but it will also join the ends of single-stranded RNA and can ligate the 3′ hydroxyl of RNA to the 5′ phosphate of DNA in a double stranded form. The enzyme requires ATP unless the 5’ end of the substrate is preadenylated.
